    ANTIBODY RESPONSES TO ANTIGENIC DETERMINANTS OF INFLUENZA VIRUS HEMAGGLUTININ : I. Thymus Dependence of Antibody Formation and Thymus Independence of Immunological Memory

    Using immunodiffusion methods it has been shown that purified hemagglutinin (HA) extracted from two related strains of influenza A viruses (A/PR8/34 and A/FM1/47) have two distinct antigenic determinants, or groups of determinants. One determinant is cross-reactive while the other is strain-specific. Antisera raised in normal mice against HA were shown to contain two populations of antibody molecules, each directed against one of the determinants. Immunization of thymus-deprived (TXBM) mice showed a strong thymus dependence of antibody formation to HA. However, the thymus dependence of antibody formation against the cross-reactive determinant could be overcome by repeated inoculations of HA in TXBM mice, indicating a different handling of two portions of the same molecule by the immunological system. Strong, secondary-type responses to the strain-specific determinant were observed in primed thymus-deprived mice after reconstitution with virgin thymus cells, showing that specific immunological memory was elicited by this determinant despite the absence of detectable antibody secretion. These findings are interpreted as examples of immunological recognition and memory mediated by B lymphocytes and discussed in terms of mechanisms of T and B lymphocyte co-operation. It is suggested that the helper effect of T lymphocytes is exerted at a late stage in the differentiation of specific populations of B cells into antibody-secreting cells

    Paper: Expert Viewers\u27 Preferences for Higher Frame Rate 3D Film

    Recently the movie industry has been advocating the use of frame rates significantly higher than the traditional 24 frames per second. This higher frame rate theoretically improves the quality of motion portrayed in movies, and helps avoid motion blur, judder and other undesirable artifacts. Previously we reported that young adult audiences showed a clear preference for higher frame rates, particularly when contrasting 24 fps with 48 or 60 fps. We found little impact of shutter angle (frame exposure time) on viewers’ choices. In the current study we replicated this experiment with an audience composed of imaging professionals who work in the film and display industry who assess image quality as an aspect of their everyday occupation. These viewers were also on average older and thus could be expected to have attachments to the “film look” both through experience and training. We used stereoscopic 3D content, filmed and projected at multiple frame rates (24, 48 and 60 fps), with shutter angles ranging from 90◦ to 358◦, to evaluate viewer preferences. In paired-comparison experiments we assessed preferences along a set of five attributes (realism, motion smoothness, blur/clarity, quality of depth and overall preference). As with the young adults in the earlier study, the expert viewers showed a clear preference for higher frame rates, particularly when contrasting 24 fps with 48 or 60 fps. We found little impact of shutter angle on viewers’ choices, with the exception of one clip at 48 fps where there was a preference for larger shutter angle. However, this preference was found for the most dynamic “warrior” clip in the experts but in the slower moving “picnic” clip for the naïve viewers.     Evidence that Viewers Prefer Higher Frame Rate Film

    High frame rate (HFR) movie-making refers to the capture and projection of movies at frame rates several times higher than the traditional 24 frames per second. This higher frame rate theoretically improves the quality of motion portrayed in movies, and helps avoid motion blur, judder and other undesirable artefacts. However, there is considerable debate in the cinema industry regarding the acceptance of HFR content given anecdotal reports of hyper-realistic imagery that reveals too much set and costume detail. Despite the potential theoretical advantages, there has been little empirical investigation of the impact of high-frame rate techniques on the viewer experience. In this study we use stereoscopic 3D content, filmed and projected at multiple frame rates (24, 48 and 60 fps), with shutter angles ranging from 90 degrees to 358 degrees, to evaluate viewer preferences. In a paired-comparison paradigm we assessed preferences along a set of five attributes (realism, motion smoothness, blur/clarity, quality of depth and overall preference). The resulting data show a clear preference for higher frame rates, particularly when contrasting 24 fps with 48 or 60 fps. We found little impact of shutter angle on viewers’ choices, with the exception of one measure (motion smoothness) for one clip type.     Seasonal changes in water quality and Sargassum biomass in southwest Australia

    Sargassum C. Agardh is one of the most diverse genera of marine macro-algae and commonly inhabits shallow tropical and sub-tropical waters. This study aimed to investigate the effect of seasonality and the associated water quality changes on the distribution, canopy cover, mean thallus length and the biomass of Sargassum beds around Point Peron, Shoalwater Islands Marine Park, Southwest Australia. Samples of Sargassum and seawater were collected every three months from summer 2012 to summer 2014 from four different reef zones. A combination of in situ observations and WorldView-2 satellite remote-sensing images were used to map the spatial distribution of Sargassum beds and other associated benthic habitats. The results demonstrated a strong seasonal variation in the environmental parameters, canopy cover, mean thallus length, and biomass of Sargassum, which were significantly (P < 0.05) influenced by the nutrient concentration (PO43-, NO3-, NH4+) and rainfall. However, no variation in any studied parameter was observed among the four reef zones. The highest Sargassum biomass peaks occurred between late spring and early summer (from September to January). The results provide essential information to guide effective conservation and management, as well as sustainable utilisation of this coastal marine renewable resource

    Splenotoxic effect of radiographic developer effluent on Wistar rats

    Background: Histological changes associated with toxicity of radiographic developer effluents on spleen tissues have not been previously studied. The present study therefore aimed at demonstrating the histopathological changes in splenic tissues of Wistar rats following exposure to developer effluent.Methods: Eighteen young Wistar rats weighing 140-220g were used for the study. The animals were divided randomly into three groups of 6 rats each based on the dose of developer effluent administered to them – i.e. control group I (0 dose) and experimental groups II (lower dose, 200 mg/kg) and III (higher dose, 400 mg/kg) respectively. The groups were further classified as either A or B sub-groups of three rats each, depending on the duration (14 or 28 days) of effluent administration. The effluent administration was done by oral gavages.Results: Normal spleen histology was observed in the control group. In contrast, tissue degeneration and necrosis; lymphocytic infiltration as well as reduction of splenic follicles were observed in some of the test groups (IIA, IIB and IIIA). Interestingly, the toxic effects of the developer effluent on group IIIB administered with higher dose for a longer period of 28 days were not as severe as observed in the other test groups.Conclusions: The present study which indicated adverse effects of exposures to sub-lethal doses of developer effluent on Wistar rats’ spleen tissues suggests the need for proper management and disposal of radiographic effluents.

    Cerebral monitoring with transcranial Doppler ultrasonography improves neurologic outcome during repairs of acute type A aortic dissection

    ObjectiveNeurologic complications after repair of acute type A aortic dissection remain significant. The use of power M-mode transcranial Doppler monitoring to verify cerebral blood flow during these repairs might decrease cerebral ischemia by correcting malperfusion. The purpose of this study was to analyze the use of power M-mode transcranial Doppler monitoring during repairs of acute type A dissection with regard to neurologic outcome.MethodsWe performed a prospective study of patients undergoing repairs of acute type A aortic dissection. Repairs included profound hypothermic circulatory arrest and retrograde cerebral perfusion. Patients in whom transcranial Doppler monitoring was used to monitor cerebral blood flow and modify operative technique during repair (study group) were compared with those without monitoring and modification (control group).ResultsBetween September 2001 and October 2003, we repaired 56 cases of acute type A dissection. Power M-mode transcranial Doppler monitoring was used in 50% (28/56) of cases. Power M-mode transcranial Doppler monitoring altered operative cannulation and guided retrograde cerebral perfusion flow in 28.5% (8/28) and 78.6% (22/28) of cases, respectively. Two patients presented with preoperative stroke, one in each group. One operative death occurred in each group. In-hospital mortality and the occurrence of new stroke were not significantly different between the 2 groups. Temporary neurologic dysfunction occurred less often in the study group (14.8% [4/27] vs 51.8% [14/27], P = .008).ConclusionsIdentification of cerebral malperfusion requires cerebral monitoring. By ensuring cerebral blood flow by using power M-mode transcranial Doppler monitoring and correcting cerebral malperfusion by modifying operative technique, neurologic outcome was improved during repairs of acute type A aortic dissection

    Molecular basis of a novel adaptation to hypoxic-hypercapnia in a strictly fossorial mole

    <p>Abstract</p> <p>Background</p> <p>Elevated blood O<sub>2 </sub>affinity enhances survival at low O<sub>2 </sub>pressures, and is perhaps the best known and most broadly accepted evolutionary adjustment of terrestrial vertebrates to environmental hypoxia. This phenotype arises by increasing the intrinsic O<sub>2 </sub>affinity of the hemoglobin (Hb) molecule, by decreasing the intracellular concentration of allosteric effectors (e.g., 2,3-diphosphoglycerate; DPG), or by suppressing the sensitivity of Hb to these physiological cofactors.</p> <p>Results</p> <p>Here we report that strictly fossorial eastern moles (<it>Scalopus aquaticus</it>) have evolved a low O<sub>2 </sub>affinity, DPG-insensitive Hb - contrary to expectations for a mammalian species that is adapted to the chronic hypoxia and hypercapnia of subterranean burrow systems. Molecular modelling indicates that this functional shift is principally attributable to a single charge altering amino acid substitution in the β-type δ-globin chain (δ136Gly→Glu) of this species that perturbs electrostatic interactions between the dimer subunits via formation of an intra-chain salt-bridge with δ82Lys. However, this replacement also abolishes key binding sites for the red blood cell effectors Cl<sup>-</sup>, lactate and DPG (the latter of which is virtually absent from the red cells of this species) at δ82Lys, thereby markedly reducing competition for carbamate formation (CO<sub>2 </sub>binding) at the δ-chain N-termini.</p> <p>Conclusions</p> <p>We propose this Hb phenotype illustrates a novel mechanism for adaptively elevating the CO<sub>2 </sub>carrying capacity of eastern mole blood during burst tunnelling activities associated with subterranean habitation.</p
